چکیده

ROTHMAN, S. S., AND F. P. BROOKS. Electrolyte secretion from rabbit pancreas in vitro. Am. J. Physiol. 208(6) : I 171-1176. 1965. -A technic has been developed permitting direct collection of undiluted rabbit pancreatic secretion in vitro without vascular perfusion. The rates of secretion and output of electrolytes were comparable to those obtained in situ. When secretin was added, flow and bicarbonate concentration increased. Secretion in vitro ceased when inhibitors of glycolysis and aerobic metabolism were added to the bathing solution. The sodium concentration of the secretion exceeded that of the bathing solution in all but one observation. Potassium concentrations in secretion were linearly related (slope = 0.93) to the potassium concentration in the bath over a range of 5.8-12.0 mM. After 4-5 hr in vitro, the total carbon dioxide concentration of secretion had decreased while chloride concentration increased with no significant change in the rate of secretion. When secretory rate changed over a range of approximately 30-600 pliters/hr, chloride and carbon dioxide output varied directly with the rate of secretion. The osmolarity of the bathing solution and secretion were always equivalent. These results are incompatible with direct plasma filtration and bicarbonate-chloride exchange as the main mechanisms of pancreatic secretion.

Micropuncture analysis of the cellular mechanisms of electrolyte secretion by the in vitro rabbit pancreas

Micropuncture techniques have been used to study electrolyte secretion by the spontaneously secreting in vitro rabbit pancreas over a wide range of environmental conditions. Pancreatic secretion does not have a strong requirement for HCO3 and secretion continues at nearly normal rates when exogenous HCO3 is replaced by acetate. Acetate concentration in the juice averages 70 meq/liter, nearly th...

A Micropuncture Investigation of the Whole Tissue Mechanism of Electrolyte Secretion by the In Vitro Rabbit Pancreas

Micropuncture techniques have been used to examine electrolyte secretion by the in vitro rabbit pancreas. The concentration profiles of the major secreted ions and digestive protein and the electrical potential profile within the pancreatic ductal system have been determined during spontaneous and secretin-stimulated secretion. The active transport of both Na and HCO(3) are the rate-controlling...

Physiol. 225(4): 781-787. 1973.-Purified cholera toxin added to the mucosal solution of rabbit ileum in vitro caused changes in ion fluxes and electrical properties after a 50to 60-min lag period. Potential difference and short-circuit current increased an...

Regulation of insulin secretion studied with pieces of rabbit pancreas incubated in vitro.

The purpose of the present study was to develop a simple system in vitro suitable for studies on the control of insulin secretion and based on the rate of release of insulin into the incubation medium from pieces of rabbit pancreas. Rabbit pancreas was chosen for these studies because it is thin (thus permitting ready diffusion of gases and other substances) and because it has a high concentrat...

Some physiological observations on the exocrine pancreas. The effects of some agents on pancreatic secretion.

Many problems are left unsolved on the exocrine pancreas. Pancreatic secretion is controlled by various factors while maintaining close relations with neighbouring organs. From the view point that the stomach and the pancreas are closely associated by way of gastrointestinal hormones, the factors involved in the mechanism of pancreatic secretion were investigated experimentally and clinically b...
